Topical Steroid Overuse Shows Skin Effects but No Ovarian Impact in Mice

Glucocorticoids are common drugs in dermatology because they calm inflammation and suppress the immune system. Many earlier experiments have shown that when these hormones enter the body—whether from stress or from taking them by mouth—they can speed up aging in the ovaries. However, it is still unclear whether simply applying a strong steroid cream to the skin can have the same effect.

The team used female C57BL/6 mice, six in each group, and applied a very high dose of fluocinolone acetonide to the skin. This supratherapeutic regimen caused the outer and inner layers of the skin to thin and the skin’s protective barrier to weaken. Researchers checked the damage with histology and by measuring barrier markers.

After the treatment, the mice’s menstrual‑like cycles stayed regular. Counting the tiny follicles in the ovaries showed no loss compared with controls. The tissue did not become more fibrous, and levels of AMH, p16INK4a and γH2AX remained unchanged. In short, the ovaries looked normal under the conditions tested.

The experiment did not track how much of the steroid entered the bloodstream, so systemic exposure is unknown. Only six mice per group were used, which limits the ability to spot small effects. Because of these gaps, the results cannot guarantee ovarian safety, and larger, longer trials are needed.
